Well... Just poking fun at Miku's surname "Hatsune", meaning "first sound". At least, a decent Vocaloid fan should know that this doesn't mean that she's the first Vocaloid, so this comic is more of a humor in purpose than anything else.
...And if we want to stretch it a bit...
Lola: I'm the first female Vocaloid ever!
I'm the first male Japanese Vocaloid!
But well, the "first sound" is really meaningful, though; it refers to her being the first Japanese Vocaloid in the Vocaloid2 engine. Overall, she's the seventh Vocaloid.

To be fair thought Miku, was the first Japanese vocaloid on the V2 engine(witch you already mentioned), the first in crypton's character vocal series and the first to become popular so she was the first in some ways (although not is the ways you would think at first.)

Actually, Leon and Lola were released at the same time so they're both the first vocaloids, female and male.
But I believe Miku's name has something to her being the first in the CV-series. With her CV01.
Kaito and Meiko's product numbers is CRV1 and CRV2. Sweet Ann, Leon, Lola, and Miriam isn't even Crypton, so I believe they thought of her as the first japanese Vocaloid 2 too.
