I make my avatars on Photoshop, with the Pencil tool (on Size 1) - I draw them pixel-by-pixel. c:

For the blinking action, I made several different versions - each with the eyes closed a little more each time. I then strung them together in an animation using Adobe Imageready (which is nowadays built-in to Photoshop). c:
