When it comes to constructing a solid character design, every detail counts. Everything from the color to the expression to the body language to the angle of the pose affect the viewer's perception, and the amount of thought put into each is difficult to hide. When it comes to this particular character design, attempting to hide any of it would be criminal.
The most immediate detail of note revolves around the mouth. The way you curved the snout, added the fangs, angled the head, and positioned the tongue when combined with the overwhelming green tone of the body immediately brings serpent-like images to mind, and the eyes and eyebrows support the original expression. Immediately and intuitively, I have a mental image of this character's base personality.
The face is supported by the fancy hairstyle she's showing off, which blends in with her ears to the point of essentially making the ears further hair decorations. Given the assortment of decs and effort spent in doing up the hair in such a style, it suggests a certain level of vanity in the character. At the very least, the character wants to outwardly present a certain type of image, and an edgy one from the looks of it. Now I already have some idea of the character's mentality.
Her design is supported by the innocent-looking collar she wears around her neck, bearing a nondescript cartoon kitten on it as well as some flowery frills. The collar offsets the edgy look set by her sinister expression and wild hair, suggesting a tamer side underneath a harsh exterior, or perhaps the ability to feign innocence for her own gain. The innocence is not entirely believable due to the coloring of the collar, as the dark purple used makes it the darkest area of her entire design. Similarly, most of the hair decs and the lollipop portray that same sense of innocence in the character, with the fangs and skull clips ready to cast a shadow of doubt on the idea. Now I've picked up on a sense of duality within the character, providing an internal source of conflict. This character is already interesting just from noting these three aspects.
While the character presented in the design is quite impressive, the more technical aspects manage to match it. This picture does a phenomenal job at varying its line width and color to create an interesting design. Most of the colors fall into the yellow-green portion of the spectrum, with each of the main elements that provide intrigue to her character dipping into blues, purples, and pinks. The color palette remains toward the middle of the color spectrum with some bits dipping to the cool side. The color usage is clearly calculated, as there is no presence of red or orange to be found, keeping the color balance in check.
Between the excellent shapes of the mouth and the eyes, the expression is wonderfully captivating. The way she looks off into the distance implies that her mind is occupied with other matters. In other words, she's scheming something, and it reflects on the rest of her physical appearance. I adore the way you layered your shapes to form the shape of the hair. It flows around her head like a flowing skirt or a pair of cheerleader pom-poms, and the bulb sticking out of the top gives it more of an asymmetrical edge that makes her head look like a music note and fits with a more antagonistic design. I also like the subtle yellow-white outline surrounding the character's main outline. It's a nice artistic touch that adds an additional "highlight" that draws more attention to the character over the background.
If there's one thing that holds this image back, it's the point at which it got cut off. From the looks of it, this pony is packing some wings, and she's got some yellow thing on her hoof. The body language is solid, but it looks like there is still more for it to say. The exact cutoff point is a little awkward, though I can see that more than the head was required to capture the realism of her holding the lollipop with her hoof. It's also clear that the background wasn't the focus here, so it could no doubt be improved as well. The pink background doesn't clash, though it might be throwing off the color balance a little given the overabundance of yellows and greens.
This picture is well-drawn, well-planned, and well-executed. The character appeal leaps out holding a megaphone, and the personality of the character is right underneath the spotlight. This is a fantastic character design, maybe one of the best I've ever seen from MLP fanart. It hits the right balance of simple and detailed. I have little to suggest in the way of improvement.

You are very welcome. I usually mention this at the beginning, but I left it out since it was a critique. But your picture was the one that most stood out from yesterday's Drawfriend on EquestriaDaily. I always see so many great works of art from the Drawfriend get so very few comments, and providing that extra bit of constructive feedback is something I'm capable of doing. I recently started picking out one image from it per day to analyze. I've been very impressed with all the ones I've seen so far. Most of them excel in form, coloring, technique, perspective, and (if an original character) design. Generally, there's one trait that stands out as most impressive, and your character design for this piece fits that bill. Not only is this a character I could see being in the show, but I honestly felt a sense of excitement just from the prospect of this character appearing in the show, knowing only the information provided from this single image. This is a powerful design, and it's no doubt bolstered by the skill in other factors offering their support.
This is one you should feel good about. This is one for your portfolio.
