I gave up on trying to understand Doctor Who a long time ago. My line is, "it's Doctor Who, it's not supposed to make sense!" Trying to make sense of it all just makes my head hurt. Besides, the total nonsense of the show is part of what makes it fun!

I don't get the question about River, her existence doesn't make less sense than other people's existence in two different universes.

Well the way I see it in the first universe Rory is dead/an auton, and also in any case it ended before anything could happen, there was no future for River to be conceived in. She was only conceived in the second universe, as a direct result of the first one being destroyed.

Why wouldn't she be able to exist/travel in different universe like The Doctor and his companions, though? It was stated that she was a complicated time event, we could assume that she doesn't quite follow the rules of normal linear people.
It makes sense in the show's logic, not with real life concepts of course.

I think the regeneration-disabled thing was just a feature of the particular poison. O_o Idk. I made up a disease that did that once...

They did briefy explain it. The scene when the Doctor is in the Tardis talking to a voice interface and it ends up being Amelia, he says after some banter "So you're saying regenerate?" and then Amelia said something snarkey about him not being able to regenerate because of the poision had some special power or something. It was said I just can't remember it word for word.
Doctor Who is a very confusing and detail focused show so it's important to pay attention.

Yeah it seems like they would have explained why his regenerations were disabled. I hope it's part of a story arc and not just a convenient plot device!

It seems pretty obvious that an assassin would use a poison that overcame the ability to regenerate if she was going to kill somebody who had that ability surely? A plot point so self-evident it needed no explanation really.
